96 Well Plate or 40 Vial Rack?

The VROC® initium one plus is the leading automatic viscometer that measures true viscosity with the highest accuracy and repeatability. Utilizing RheoSense's patented Viscometer/ Rheometer-on-a-Chip (VROC) technology, RheoSense's viscometers (combining MEMS and microfluidics) have redefined the viscometry industry, offering the smallest sample volume per measurement coupled with exceptional ease-of-use and accuracy. Superior resolution makes it possible to measure hydrodynamic size (Intrinsic Viscosity) and to quantify protein-protein interactions (PPI) and protein stability with the VROC initium one plus.

The VROC initium one plus includes an intuitive software that allows automatic testing of samples on a 96 well plate or a 40 vial rack. With this automation you can "set and forget" your viscosity measurements, greatly increasing efficiency and allowing your experts to focus their time and attention on their most valuable work.

While the VROC initium one plus can be programmed to run samples on both a 96 well plate and a 40 vial rack,  different groups tend to only use one or the other. For example, when working in early stage drug discovery, groups often work with limited samples and don't have as many samples to test, making the 40 vial rack is a better option for sample testing. The vials used in the 40 vial rack are HPLC vials, making them much easier to load (than the 96 well plate). Most groups are already very familiar with 40 vial racks and often already have them in their laboratory. This eliminates the need to purchase different vials, as HPLC vials are compatible. VROC initium one plus 40 vial rack vials can be loaded with 26-80µL of your sample. This minimal sample requirement, coupled with the advanced retrieval and recovery feature of the VROC initium one plus allows for unlimited testing on the same sample. Vial #5

Groups that have a lot of screening tend to utilize the 96 well plate. Groups already working with an autoloader are especially drawn to the 96 well plate as most autoloaders are set up to load 96 well plate. It can be difficult and time consuming to load a 96 well plate. Using an autoloader provides a seamless transition from autoloader to VROC initium one plus. It is recommended that the VROC initium one plus 96 well plate be loaded with ~35µL of sample. image-png-Dec-07-2021-06-17-47-13-PM

Sealing films are required for the 96 well plate to prevent evaporation and contamination. To be compatible with the autosampler syringe used for loading, the film must be easily punctured without coring or fracturing which could lead to clogging along the flow path or in the microfluidic channel. Many films are not intended to be pierced and lead to failure or damage. Although seals made of foil are easily punctured, they can produce deformable fragments that can be introduced into the flow path. We have identified a polytetrafluoroethylene (PTFE) film designed to be punctured without coring or needle clogging (J.G. Finneran Associates, Inc). The PTFE film is compatible with aqueous solutions and expected to have excellent chemical resistance to a broad range of organic solvents. To assess the performance of the PTFE sealing film, an extensive test was conducted including 30 samples of a concentrated model protein solution. We monitored for consistent puncturing of the film, loaded volume, and viscosity value.
